With the pulse dose method the fish are first fed a discrete marker meal in the morning, when they are hungry. After the whole meal has been eaten they are fed normal feed and faeces is sampled at short intervals as long as the marker appears in the faeces. Because the faeces subsamples are very small this is only possible if reliable micro-analysis methods to quantity the marker concentration are available.
Based on the time series of faeces samples in the microgram range, an excretion curve is constructed, where the marker concentration decreases non-linearly with time. The faecal output can then be calculated from the area under curve. When the indigestibility has been quantifyed using the faecal dilution method, this can be combined with the faecal output as an estimate of feed intake. Because this method was developed with sheep it is still uncertain if it works in fish nutrition research. |
